Cardiometabolic Safety of Lumateperone (ITI−007): Post Hoc Analyses of Short-Term Randomized Trials and an Open-Label Long-Term Study in Schizophrenia

Study Objective
Current treatments for schizophrenia are often associated with increased rates of metabolic syndrome (MetSy). MetSy is defined as meeting 3 of the following 5 criteria: waist circumference >40in (men) or >35in (women), triglycerides =150mg/dL, high density lipoprotein cholesterol (HDL) <40mg/dL (men) or <50mg/dL (women), systolic blood pressure (BP) =130mmHg or diastolic BP =85mmHg, fasting glucose =100mg/dL. Patients with MetSy have an elevated risk of developing type II diabetes and increased mortality due to cardiovascular disease. Lumateperone (lumateperone tosylate, ITI−007), a mechanistically novel antipsychotic that simultaneously modulates serotonin, dopamine, and glutamate neurotransmission, is FDA approved for the treatment of schizophrenia. This distinct pharmacological profile has been associated with favorable tolerability and a low risk of adverse metabolic effects in clinical trials. This post hoc analysis of 2 randomized, double-blind, placebo-controlled studies of patients with an acute exacerbation of schizophrenia compared rates of MetSy with lumateperone and risperidone. Data from an open-label long-term trial of lumateperone were also evaluated.
MethodThe incidence and shift in MetSy were analyzed in data pooled from 2 short-term (4 or 6 week) placebo- and active-controlled (risperidone 4mg) studies of lumateperone 42mg (Studies 005 and 302). The pooled lumateperone data were compared with data for risperidone. Data from an open-label 1-year trial (Study 303) evaluated MetSy in patients with stable schizophrenia switched from prior antipsychotic (PA) treatment to lumateperone 42mg.
ResultsIn the acute studies (n=256 lumateperone 42mg, n=255 risperidone 4mg), rates of MetSy were similar between groups at baseline (16% lumateperone, 19% risperidone). At the end of treatment (EOT), MetSy was less common with lumateperone than with risperidone (13% vs 25%). More lumateperone patients (46%) compared with risperidone (25%) patients improved from having MetSy at baseline to no longer meeting MetSy criteria at EOT. Conversely, more patients on risperidone than on lumateperone developed MetSy during treatment (13% vs 5%). Differences in MetSy conversion rates were driven by changes in triglycerides and glucose. In the long-term study (n=602 lumateperone 42mg), 33% of patients had MetSy at PA baseline. Thirty-six percent of patients (36%) with MetSy at PA baseline improved to no longer meeting criteria at EOT. Fewer than half that percentage shifted from not meeting MetSy criteria to having MetSy (15%).
ConclusionsIn this post hoc analysis, lumateperone 42mg patients had reduced rates of MetSy compared with risperidone patients. In the long-term study, patients with MetSy on PA switched to lumateperone 42mg had a reduction in the risk of MetSy. These results suggest that lumateperone 42mg is a promising new treatment for schizophrenia with a favorable metabolic profile.

This article reports the structure and morphology of the in vivo interface between implants composed of either α tricalcium phosphate (αTCP) or αTCP doped with 3.0 wt% dicalcium silicate (αTCPss) ceramic, and natural bone of rabbit tibias. Both interfaces developed a new bone layer in direct contact with the implants after 4 and 8 weeks of implantation. The specimens were examined using analytical scanning and transmission electron microscopy, up to the lattice plane resolution level. Degradation processes of the implants developed at the interfaces encouraged osseous tissue ingrowth into the periphery of the material, changing the microstructure of the implants. The ionic exchange initiated at the implant interface with the environment was essential in the integration process of the implant, through a dissolution–precipitation–transformation mechanism. The interfaces developed normal biological and chemical activities and remained reactive over the 8-week period. Organized collagen fibrils were found at the αTCPss/bone interface after 4 weeks, whereas a collagen-free layer was present around the Si-free αTCP implants. These findings suggest that the incorporation of silicate ions into αTCP ceramic promotes processes of the bone remodeling at the bone/αTCPss interface, hence the solubility rate of the αTCPss material decreased.
Pb(Po)-210 concentration of tobacco samples grown in the vicinity of a remedied uranium mine

The environmental monitoring of waste heaps, sludge reservoirs is an important task. The Pb(Po) accumulation of tobacco has been known for long. Knowing familiar with the accumulation capability can be suitable for bio-monitoring the tailing ponds of the remedied uranium mine, as isotopes may be bound on the skin surfaces or may be absorbed via the roots.
Our research investigates the Pb(Po)-210 concentration values of tobacco plants grown for experimental purposes in the remedied uranium-mine area. During the work the Pb(Po)-210 concentration values of 73 tobacco and soil samples, grown between 2002-2009 were determined using a semi-conductor (PIPS) detector alpha-spectrometer. The source for measurement was prepared using spontaneous deposition process following the combined acidic leaching using Po-209 tracers. Based on the results it can be stated that the Pb(Po)-210 activity concentration values of tobacco and soil samples altered between 2.12 ± 0.8 – 1866 ± 98 mBq/g. Processing the measurement data of the previous years it was apparent that with the changes of the Pb(Po) concentration values of the soil samples the activity concentration values of the tobacco samples also changed proportionally. Therefore, later the migration of radioisotopes disengaged from the tailings dump can also be traced.

The thermomechanical behavior of Cu thin films, 600–1125 nm thick and encapsulated between SiNx barrier and SiNx or AlNx passivation layers on silicon substrates, was studied during thermal cycling between room temperature and 400 or 500 °C using the substrate curvature method. Films were prepared with varying oxygen contents, and the distribution of oxygen through the thickness of selected films was studied before and after thermal cycling using secondary ion mass spectrometry. Large variations in the thermomechanical behavior with oxygen content were found and correlated with segregation of oxygen to the film/barrier and film/passivation interfaces. These variations are thought to be due to recovery of stored misfit dislocation energy, which is, in turn, controlled by oxygen in the film. Effects of oxygen on film deformation through variations in interfacial adhesion and diffusion-induced dislocation glide are considered.

A comprehensive study of a steady axisymmetric supersonic jet of CO2, including experiment, theory, and numerical calculation, is presented. The experimental part, based on high-sensitivity Raman spectroscopy mapping, provides absolute density and rotational temperature maps covering the significant regions of the jet: the zone of silence, barrel shock, Mach disk, and subsonic region beyond the Mach disk. The interpretation is based on the quasi-gasdynamic (QGD) system of equations, and its generalization (QGDR) considering the translational–rotational breakdown of thermal equilibrium. QGD and QGDR systems of equations are solved numerically in terms of a finite-difference algorithm with the steady state attained as the limit of a time-evolving process. Numerical results show a good global agreement with experiment, and provide information on those quantities not measured in the experiment, like velocity field, Mach numbers, and pressures. According to the calculation the subsonic part of the jet, downstream of the Mach disk, encloses a low-velocity recirculation vortex ring.
Environmentally Stable, Low Dielectric Polymers from Soluble Intermediates

We have investigated new polymers of poly(phenylene vinylene), PPV and poly(phenylene), PP. Like polyimides, these polymers can be spin-coated from soluble intermediates and converted to their thermally stable forms by elimination of small groups during a heating cycle. As such, these polymers are insulating. Both poly(phenylene vinylene) and poly(phenylene) also have the potential for metallic conductivity when properly doped. During curing, the complete removal of the leaving groups is important in determining the final behavior of the aromatized polymer film. Elimination of the leaving groups was followed by means of Rutherford Backscattering (RBS) and Forward Recoil Spectrometry (FRES) analysis for various curing conditions and environments. We will describe the curing and film-forming properties of these polymers, and discuss both their dielectric characteristics and thermal stability as a function of the curing conditions.
The Pit Alignment at Eskbank Nurseries

Eskbank Nurseries, a market garden allotment some 200 m long and 30 to 40 m wide, lies at approximately 70 m OD on a relatively level terrace overlooking the south bank of the river North Esk (fig. 1). The excavated area is centred on NT320660 and lies within the two superimposed Roman camps first noted as crop marks by St Joseph in 1965 and subsequently excavated by Maxfield in 1972. Her excavation areas I and II were sited to investigate a pit alignment, visible on the aerial photographs and plotted by her (Maxfield 1974, 142, fig. 1). In the event no pits were discovered by excavation, a fact attributable to an unfavourable combination of weather and soil conditions (Maxfield 1974, 150). The pit alignment did not, of course, appear as a crop mark in the deep market-garden soils of the Nurseries but, by extrapolation of Maxfield's plot, it was clear that it must cross the northern half of the allotment, possibly intersecting the north ditch of the earlier of the Roman camps (Maxfield's Camp A) on the west edge of the site.
